ABSTRACT

Objective:The non targeted high-throughput urine metabolomics technology was used to study the pathogenesis of APP/PS 1 double transgenic mice and the mechanism of action of Gouteng san.Methods:5-month-old APP/PS 1 double transgenic mice were test with Morris water maze for spatial learning ability.Then we employed the non targeted high-throughput urine metabolomics technology to study the pathogenesis of APP/PS1 double transgenic mice based on the metabolic network.The focus investigation of the key pathways and the observation of the treatment by Morris water maze and metabolic level have been used after spatial learning ability damaged confirmed.Results:The comparison between APP/PS1 double transgenic mice and normal mice suggested that a significant longer was existed in former,which was call-back by Gouteng san.With the non targeted high-throughput urine metabolomics analysis and pathway focused analysis,we found certain signals from metabolic profiling,which was identified to be 6 biomarkers associated with learning and memory function by mass spectrometry analysis or authoritative database.Respectively,they were taurine,pteroylglutamic acid,neopterin,glutaurine,2-oxoglutarate and dihydroneopterin.They were mainly related to taurine metabolism and folate metabolism and represented an effective callback.Conclusion:Gouteng san possess a favorable effect on learning and memory ability of APP/PS1 double transgenic mice,6 biomarkers may be a potential target for the pathogenesis of APP/PSI double transgenic mice and provide experimental basis for the study of Gouteng san.

ABSTRACT

Objective To analyze the causes for the failures after posterior pedicle screw instrumentation for thoracolumbar fractures.Methods From June 2003 to December 2014,182 patients with thoracolumbar fracture were treated by fixation through the posterior approach using pedicle screws and fully followed up in our institute.We analyzed the cases of postoperative infection,recovery of neural symptoms,breakage and loosening of pedicle screws and connecting rod,non-union of the fractured vertebra,and correction loss of kyphosis in associations with the AO classification and Loading Sharing Classification of Spine Fracture (LSCSF) system,osteoporosis,intervertebral disc injury and methods of internal fixation.Results In this series,altogether 27 cases failed(14.8%).The rate of postoperative infection was 1.1% (2/182).The rate of breakage of pedicle screw or connecting rod was 7.7% (14/182).The implant breakage rates for fractures of AO types A1,A2 and A3.1 were significantly lower than for other types (P < 0.05).The implant breakage rate for the patients with ≤6 LSCSF points was significantly lower than for those with ≥7 LSCSF points (P < 0.05).The implant breakage occurred in 3 cases of those who underwent fixation of one normal vertebra respectively below and above the two contiguous segments but not in those who underwent additional fixation of the injured vertebrae.The rate of screw loosening was 2.2% (4/182).The non-union rate of the injured vertebra was 2.7% (5/182).The rate of kyphosis recurrence was 1.1% (2/182).Conclusions To prevent the failure of posterior pedicle screw fixation,surgeons should pay more attention to the following key points before operation:the type and evaluation of spinal fractures,a proper approach and method of internal fixation,and the weight bearing capability of the anterior column.

ABSTRACT

From the perspective of portable monitoring devices,we use an analog front-end AFE4490 design a module of Non-invasive blood oxygen measurement, used to collect human pulse wave signal and peak (valley) value detection and then use the principles of non-invasive oximetry calculated oxygen saturation (SPO2). This design of noninvasive oximetry module has the characteristics of small size, low power consumption, and the results of test show that the measurement of oxygen saturation are correct.

ABSTRACT

Recently, in situ protein microarrays have been developed for large scale analysis and high throughput studies of proteins. In situ protein microarrays produce proteins directly on the solid surface from pre-arrayed DNA or RNA. The advances in in situ protein microarrays are exemplified by the ease of cDNA cloning and cell free protein expression. These technologies can evaluate, validate and monitor protein in a cost effective manner and address the issue of a high quality protein supply to use in the array. Here we review the importance of recently employed methods: PISA (protein in situ array), DAPA (DNA array to protein array), NAPPA (nucleic acid programmable protein array) and TUSTER microarrays and the role of these methods in proteomics.

ABSTRACT

Objective To observe the relationship between the change of endothelin-1 (ET-1), nitric oxide synthase(NOS) content and cerebral vasospasm after experiment Sprague-Dawley rat subarachnoid hemorrhage (SAH). Methods SD rat got the tail artery blood and followed prone positioning in a stereotaxic apparatus, injected autologous arterial blood or saline into cisterna magna,2 d later injected in the same way. For basilar artery (BA),internal carotid artery(ICA) ,and middle cerebral artery(MCA) diameter measurements,gelatin-india ink vascular casted,the blood ET-1 and NOS content were detected by immunoradiology or colorimetry measurement. Results Microangiography in SAH showed extensive macroscopic filling deficit,the diameter of MCA,ICA and BA ((169.33 ±8.67)mm, (227.33 ± 14.25) mm, (226.33 ±5.99) mm respectively) were shorter than those in control group((259.5 ±7.48)mm,(228.17 ±8.09)mm,(254. 67 ±8.48)mm respectively) (all P<0.05). Neuronal function score of 7d(9.45 ±1.77) was higher than control group(0.60 ±0.49) (P<0.05). Content of ET-1 and NOS in blood after SAH 7 d were(231.25 ± 19.45)g/L,(198.50 ±9.52) × 103U/L) ,(72.99 ±5.83)g/L, (230.76 ±17.06) × 103U/L),for sham group,there was significant differences (P<0.05). Conclusion Experiment rat double-hemorrhage method from tail artery can makes experimental subarachnoid hemorrhage model,neurological functional score is increased, neuronal function score is associated with diameter of MCA and ET-1 content.

Objective To study the clinic effect of repairing the femoral head with the transposition of great trochanter bone flap pedicled with branch of transverse lateral circumflex femoral vascular Methods To opt the stage Ⅲ~Ⅳ cases of adult avascular necrosis of the femoral head, after cleared the pathological changes position, the transposition of greater trochanter bone flap pedicled with branch of transverse lateral circumflex femoral vascular were repaired the femoral head Results Twelve cases were subjected to a serial follow up (average 2 9 years). The function of hip joint were recover and the clinic good was seen. The excellent rate was 75 5% Conclusion The lack bone after the operation of avascular necrosis of the femoral head, appling with the transposition of greater trochanter bone flap pedicled with branch of transverse lateral circumflex femoral vascular to repair the femoral head, renew the function of hip joint Its an effective means to treat avascular necrosis of femoral head
